The Liver and Postheparin Plasma Lipolytic Activity in Dog and Man.

چکیده
The intravascular injection of heparin results in the rapid appearance in the blood of a lipolytic enzyme capable of clearing lipemic sera and hydrolyzing artificial triglyceride emulsions (1, 2). This enzyme has been termed clearing factor (3), lipoprotein lipase (4), clearing factor lipase (2), or postheparin plasma lipolytic activity (PHLA) (5). A lipolytic enzyme with characteristics nearly identical to those of PHLA has been identified in relatively high concentration in tissue extracts of heart (4), adipose tissue (6), and mammary gland (7, 8). Lower concentrations of this lipolytic enzyme have been found in tissue extracts of thoracic and abdominal viscera, including the liver (3, 4, 9). Release of PHLA from tissue sources into the circulation has been demonstrated in perfusion studies of limbs and abdominothoracic viscera (3, 10, 11). However, perfusion of the isolated rat liver with heparinized blood has been reported by several observers (10, 12-14) to result in net loss of PHLA and no release of lipolytic activity by the liver. Boberg, Carlson, and Normell (15), on the other hand, have reported recently in an abstract that the isolated perfused canine liver

A new method for the measurement of lipoprotein lipase in postheparin plasma using sodium dodecyl sulfate for the inactivation of hepatic triglyceride I ipasel
Lipoprotein lipase (LPL) and hepatic triglyceride lipase (H-TGL) are lipolytic activities found in postheparin plasma. A simple and precise method for the direct determination of LPL in postheparin plasma is described. Preincubation of this plasma (45-60 min at 26°C) with sodium dodecyl sulfate (35-50 mM) in 0.2 M Tris-HC1 buffer, pH 8.2, results in the inactivation of H-TGL, while leaving LPL ...
متن کاملPlasma decay of chylomicron remnants is not affected by heparin-stimulated plasma lipolytic activity in normal fasting man1
In an earlier study it was shown that retinyl palmitate appeared to be a satisfactory label for the core of chylomicrons and their remnants. When chylomicrons were endogenously labeled with retinyl palmitate and pulse-injected into healthy donors, retinyl palmitate was cleared from plasma by a first order process. Its fractional decay constant was very similar to the fractional catabolic rate o...

متن کاملDecreased plasma postheparin lipolytic activity in systemic lupus erythematosus.
Plasma postheparin lipolytic activity (PHLA) was measured on 50 patients with systemic lupus erythematosus (SLE). Plasma PHLA was significantly decreased in SLE patients. This decrease was most striking in the acute phase of the disease. There was a close relationship between decreased PHLA and immunologic factors indicative of the acute phase of SLE. These immunologic factors included shaggy a...
متن کاملDimeric lipoprotein lipase is bound to triglyceride-rich plasma lipoproteins.
Lipoprotein lipase hydrolyzes the triglyceride-rich core of chylomicrons and very low density lipoproteins. It is also a ligand, in vitro, for binding of lipoproteins to the low density lipoprotein receptor-related protein and may play a central role in the receptor-mediated removal of triglyceride-rich lipoproteins. The aim of the present study was to determine to which lipoprotein subclass th...
